Elsevier's "Pharmacological Research" has published an article on ivermectin this January, Ivermectin, a potential anticancer drug derived from an antiparasitic drug .

Ivermectin has powerful antitumor effects, including the inhibition of proliferation, metastasis, and angiogenic activity, in a variety of cancer cells. This may be related to the regulation of multiple signaling pathways by ivermectin through PAK1 kinase. On the other hand, ivermectin promotes programmed cancer cell death, including apoptosis, autophagy and pyroptosis. Ivermectin induces apoptosis and autophagy is mutually regulated. Interestingly, ivermectin can also inhibit tumor stem cells and reverse multidrug resistance and exerts the optimal effect when used in combination with other chemotherapy drugs.

The European Society of Cardiology's Heart Failure journal is reporting that aspirin use is associated with increased risk of heart failure in certain "at risk" categories.

Aspirin use is associated with an increased risk of HF in patients receiving aspirin with or without a previous history of CVDs. In the absence of conclusive trial evidence, our observations suggest that aspirin should be prescribed with caution in patients at risk of HF or having HF.

Increased risk of infection with SARS-CoV-2 Beta, Gamma, and Delta variant compared to Alpha variant in vaccinated individuals - a preprint on medrxiv.

Here, we analyze 28,578 sequenced SARS-CoV-2 samples from individuals with known immune status obtained through national community testing in the Netherlands from March to August 2021. We find evidence for an increased risk of infection by the Beta (B.1.351), Gamma (P.1), or Delta (B.1.617.2) variants compared to the Alpha (B.1.1.7) variant after vaccination. No clear differences were found between vaccines. However, the effect was larger in the first 14-59 days after complete vaccination compared to 60 days and longer. In contrast to vaccine-induced immunity, no increased risk for reinfection with Beta, Gamma or Delta variants relative to Alpha variant was found in individuals with infection-induced immunity.

The Relation of Toxoplasma Infection and Sexual Attraction to Fear, Danger, Pain, and Submissiveness

A cross-sectional cohort study performed on 36,564 subjects (5,087 Toxoplasma free and 741 Toxoplasma infected) showed that infected and noninfected subjects differ in their sexual behavior, fantasies, and preferences when age, health, and the size of the place where they spent childhood were controlled (F(24, 3719) = 2.800, p < .0001). In agreement with our a priori hypothesis, infected subjects are more often aroused by their own fear, danger, and sexual submission although they practice more conventional sexual activities than Toxoplasma-free subjects. We suggest that the later changes can be related to a decrease in the personality trait of novelty seeking in infected subjects, which is potentially a side effect of increased concentration of dopamine in their brain.

Obesity and Sex-Related Associations With Differential Effects of Sucralose vs Sucrose on Appetite and Reward Processing: A Randomized Crossover Trial. NPR summarizes:

"We found that females and people with obesity had greater brain reward activity" after consuming the artificial sweetener, says study author Katie Page, a physician specializing in obesity at the University of Southern California.Both groups also had a reduction in the hormone that inhibits appetite, and they ate more food after they consumed drinks with sucralose, compared with after regular sugar-sweetened drinks. In contrast, the study found males and people of healthy weight did not have an increase in either brain reward activity or hunger response, suggesting they're not affected in the same way.

A Report on Myocarditis Adverse Events in the U.S. Vaccine Adverse Events Reporting System (VAERS) in Association with COVID-19 Injectable Biological Products - describes what the title explains.

Within 8 weeks of the public offering of COVID-19 products to the 12-15-year-old age group, we found 19 times the expected number of myocarditis cases in the vaccination volunteers over background myocarditis rates for this age group. In addition, a 5-fold increase in myocarditis rate was observed subsequent to dose 2 as opposed to dose 1 in 15-year-old males.

Problems in the “Evidence” of “Evidence-Based Medicine” by Alvan RFeinstein, MD and Ralph I Horwitz, MD

The proposed practice of “evidence-based medicine,” which calls for careful clinical judgment in evaluating the “best available evidence,” should be differentiated from the special collection of data regarded as suitable evidence. Although the proposed practice does not seem new, the new collection of “best available” information has major constraints for the care of individual patients.

Derived almost exclusively from randomized trials and meta-analyses, the data do not include many types of treatments or patients seen in clinical practice; and the results show comparative efficacy of treatment for an “average” randomized patient, not for pertinent subgroups formed by such cogent clinical features as severity of symptoms, illness, co-morbidity, and other clinical nuances. The intention-to-treat analyses do not reflect important post-randomization events leading to altered treatment; and the results seldom provide suitable background data when therapy is given prophylactically rather than remedially, or when therapeutic advantages are equivocal. Randomized trial information is also seldom available for issues in etiology, diagnosis, and prognosis, and for clinical decisions that depend on pathophysiologic changes, psychosocial factors and support, personal preferences of patients, and strategies for giving comfort and reassurance.

The laudable goal of making clinical decisions based on evidence can be impaired by the restricted quality and scope of what is collected as “best available evidence.” The authoritative aura given to the collection, however, may lead to major abuses that produce inappropriate guidelines or doctrinaire dogmas for clinical practice.

A European group calling themselves Doctors for Covid Ethics is pursuing legal action through liability against members of the European parliament trying to push for vaccine passports.

On April 20, all following Members of the European Parliament were served with notices of liability, advising that they may be held personally liable for harm and death caused by implementation of a Digital Green Certificate (Vaccine Passport), to be voted upon in the European Parliament on April 28, 2021.
